Polymorphism of Beta-lactoglobulin Gene in Select Population Slovak Spotted Breed

The goal of the paper was to identify lactoglobulin gene polymorphism and analyse of genotype structure in population Slovak spotted breed. The lactoglobulin (LGB) expressed in milk and is important in the evaluation of milk production potential and butterfat and protein content. LGB is localized on bovine chromosome 11. The AA genotype of LGB is associated with higher milk yield, the BB genotype with higher fat and casein content and is more desirable for cheese making. The material involved 28 cattle. Bovine genomic DNA was isolated from sperm by commercial kit NucleoSpin Blood (Macherey-Nagel) and used in order to estimate LGB genotypes by means of PCR RFLP method with using HaeIII restriction enzyme. In the population included in the study, genotype AA (148 bp, 99 bp), genotype AB (148 bp, 99 bp, 74 bp), genotype BB (99 bp, 74 bp). There were detected homozygote genotype AA with frequency 50 %, heterozygote genotype AB with frequency 21.43 % and homozygote genotype BB with frequency 28.57 % in evaluated Slovak spotted breed. Results point out that frequency of A allele was high and was represented 60.71 %. Frequency of B allele was 39.29
